The receptor complex mediating the biological activities consists of two distinct membrane-bound glycoproteins:

IL-6 Receptor (IL-6R, gp80) is 80 kDa alpha subunit, transmembrane glycoprotein that is 449 aa long. It binds IL-6 with low affinity A signal-transducing non-ligand binding 130 kDa component(gp130) that is required for high-affinity binding complex (Kd = 50 pM)
Soluble IL-6 receptor (sIL-6R) is a 50-55 kDa ligand binding protein, derived from the extracellular part of the gp80 receptor by either proteolytic cleavage or by alternative splicing. The soluble form of IL-6R can bind its ligand and induce cellular responses by association with gp130, thus acting as an IL-6 agonist.

Dysregulated production of IL6 and its receptor are implicated in the pathogenesis of many diseases, such as multiple myeloma, autoimmune diseases and prostate cancer. Elevated IL-6 concentrations have been reported in numerous clinical disorders where they appear to orchestrate a variety of inflammatory responses.

Assay Principle

Human IL-6R / CD126 ELISA Assay Kit is a solid phase sandwich Enzyme Linked-Immuno- Sorbent Assay (ELISA). A monoclonal antibody specific for IL-6R has been coated onto the wells of the microtiter strips provided. Samples, including standards of known IL-6R concentrations and unknowns are pipetted into these wells.  During the first incubation, the IL-6R antigen and a biotinylated monoclonal antibody specific for IL-6R are simultaneously incubated.  After washing, the enzyme ( streptavidin-peroxydase ) is added. After incubation and washing to remove all the unbound enzyme, a substrate solution is added to induce a coloured reaction product. The intensity of this coloured product is directly proportional to the concentration of IL-6R present in the samples.

  1. Add 100μl of each standard,  diluted sample, control and zero (standard diluent) in duplicate to appropriate number of wells
  2. Add 50μl of diluted biotinylated anti-IL-6R to all wells
  3. Cover with a plastic plate cover and incubate at room temperature (18 to 25°C) for 1 hours
  4. Remove the cover and wash the plate.
  5. Add 100μl of Streptavidin-HRP solution into all wells
  6. Cover with a plastic plate cover and incubate at room temperature (18 to 25°C) for 30 min
  7. Repeat wash step 5.
  8. Add 100μl of ready-to-use TMB Substrate Solution into all wells
  9. Incubate in the dark for 10-20 minutes* at room temperature. Avoid direct exposure to light by wrapping the plate in aluminium foil
  10. Add 100μl of H2SO4:Stop Reagent into all wells
