Assay Background
Ocrelizumab, sold under the brand name Ocrevus, is a medication used for the treatment of multiple sclerosis (MS).It is a humanized anti-CD20 monoclonal antibody. It targets CD20 marker on B lymphocytes and hence is an immunosuppressive drug. Ocrelizumab binds to an epitope that overlaps with the epitope to which rituximab binds.
Assay Principle
The method employs the quantitative sandwich enzyme immunoassay technique. Human CD20 protein is precoated onto microwells. Samples and standards are pipetted into microwells and human Ocrelizumab present in the sample are bound. Then, Anti-Human IgG HRP conjugate is pipetted and incubated. After washing microwells in order to remove any non-specific binding, the ready to use substrate solution (TMB) is added to microwells and color develops proportionally to the amount of Ocrelizumab in the sample. Color development is then stopped by addition of stop solution. Absorbance is measured at 450 nm.
