Assay Principle

The principle of the following chemiluminescence immunoassay (LIA) test follows a two-step competitive binding scenario. Competition occurs between an unlabelled antigen (present in standards, control and patient samples) and an estrone-labelled antigen for a limited number of anti-estrone antibody binding sites on the microplate. The washing and decanting procedures remove unbound materials, and then luminescence substrate solution is added. The relative luminescence units (RLUs) are measured on a microtiter plate luminometer. The RLU values are inversely proportional to the concentration of estrone in the sample. A set of calibrators is used to plot a standard curve from which the amount of estrone in saliva samples and controls can be directly read.


SPECIMEN COLLECTION AND STORAGE
Approximately 1 mL of saliva is required per duplicate determination. Collect 2–3 mL of saliva into a clean glass tube without force or inducement and before eating, drinking or brushing the teeth. Simply rinse the mouth with water before collection and wait a few minutes to start. Do not use blood-contaminated specimens. Store samples at 4°C for up to 4 hours or at -10°C or lower if the analyses are to be done at a later date. Consider all human specimens as possible biohazardous materials and take appropriate precautions when handling.’

Assay Background


Estrone is a steroid like estriol and estradiol, belonging to the class of estrogens. The estrogens are involved in the development of female sex organs and secondary sex characteristics. Before the ovum is fertilized the main action of the estrogens is on the growth and function of the reproductive tract in order to prepare it for the fertilized ovum. During the follicular phase of the menstrual cycle the estrone level shows a slight increase. The production of estrone then increases markedly to peak at around day 13 of the cycle. The peak is of short duration and by day 16 of the cycle levels will be low. A second peak occurs at around day 21 of the cycle and if fertilization does not occur, then the production of estrone decreases. The measurement of estrone is useful for monitoring ovarian function, in predicting ovulation, in assessing the effect of therapy to induce ovulation. The measurement of estrone in saliva is an attractive alternative to plasma analysis because its non-invasive, stress-free procedure makes serial sampling easy. Some research appears to confirm that estrone concentrations in saliva approximate the unbound-estrone in plasma therefore provide a more accurate indication of endocrine activity.
