Assay Principle

The Zika Virus IgG ELISA Assay Kit is a “sandwich” ELISA is designed, developed and produced for the qualitative measurement of IgG antibodies in blood specimen. The assay utilizes the microplate-based enzyme immunoassay technique by coating highly purified antigen onto the wall of microtiter wells. Controls and blood specimen are added to microtiter wells of microplate that is coated with a highly purified Zika antigen on its wall. During the assay, Zika antibodies will be bound to the antigen coated plate after an incubation period. The unbound material is washed away and another HRP-conjugated polyclonal antibody which specifically recognizes human IgG is added for further immunoreactions. After an incubation period, the immunocomplex of “Zika viral antigen – anti Zika antibodies – HRPconjugated goat anti human IgG Tracer Antibody” is formed if Zika IgG antibodies are present in the test sample. The unbound tracer antibody and other proteins in buffer matrix are removed in the subsequent washing step. HRP conjugated tracer antibody bound to the well is then incubated with a substrate solution in a timed reaction and then measured in a spectrophotometric microplate reader. The enzymatic activity of the tracer antibody bound to Zika IgG antibodies captured on the wall of each microtiter well is directly proportional to the amount of Zika IgG antibody level in each test specimen.

Assay Background


Zika virus is a flavivirus, in the family Flaviviridae. Zika was discovered in Uganda and has spread rapidly to the Americas and the Caribbean in the past year. Zika virus is mainly spread by the bite of infected mosquitos (Aedes aegypti and Aedes albopictus). Zika virus can also be transmitted from person to person through sexual contact, by blood transfusion or contact with infected materials. The infection usually results in mild or sometimes no symptoms and is normally not life threatening. Women can transmit the virus to the fetus during pregnancy. Infection during pregnancy is deleterious to the fetus and is associated with fetal death, fetal growth restriction, and a spectrum of central nervous system abnormalities.

Assay Procedure


  1. Add 100 μL of Controls and diluted (1:100) patient samples.
  2. Cover the plate with an aluminum foil to avoid exposure to light. Incubate the plate at room temperature static for 30 minutes.
  3. Wash each well 5 times by dispensing 350 μl of working wash solution into each well and then completely aspirating the contents. Alternatively, an automated microplate washer can be used.
  4. Add 100 μL Zika virus Tracer Antibody.
  5. Cover the plate with an aluminum foil to avoid exposure to light. Incubate the plate at room temperature, static for 30 minutes.
  6. Wash each well 5 times by dispensing 350 μl of working wash solution into each well and then completely aspirating the contents. Alternatively, an automated microplate washer can be used.
  7. Add 100 μl of ELISA HRP Substrate into each of the wells.
  8. Cover the plate with an aluminum foil to avoid exposure to light. Incubate plate at room temperature static for 20 minutes.
  9. Remove the aluminum foil and plate sealer. Add 100 μl of ELISA Stop Solution into each of the wells. Mix gently.
  10. Read the absorbance at 450 nm within 10 minutes using a microplate reader.
