Assay Principle

The Rat Tff3 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid phase immunoassay specially designed to measure Rat Tff3 with a 96-well strip plate that is pre-coated with antibody specific for Tff3. The detection antibody is a biotinylated antibody specific for Tff3. The capture antibody is a monoclonal antibody from mouse, the detection antibody is a biotinylated polyclonal antibody from goat. The kit contains recombinant Rat Tff3 with immunogen: Expression system for standard: E.coli, Immunogen sequence: A23-F81. The kit is analytically validated with ready to use reagents. To measure Rat Tff3, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is substrate to HRP and will be catalyzed to produce a blue color product, which changes into yellow after adding acidic stop solution. The density; of the yellow product is linearly proportional to Rat Tff3 in the sample. Read the density of the yellow product in each well using a plate reader, and benchmark the sample wells’ readings against the standard curve to determine the concentration of Rat Tff3 in the sample.

Assay Background


Trefoil factor 3 is a protein that in humans is encoded by the TFF3 gene. It is mapped to 21q22.3. Members of the trefoil family can interact with mucins and have an influence on mucus viscosity. They also promote migration of epithelial cells, are linked to antiapoptosis, induce cell scattering, trigger chemotaxis, and participate in immune responses. This gene is a marker of columnar epithelium and is expressed in a variety of tissues including goblet cells of the intestines and colon. It has been found that TFF3 is the most highly upregulated gene in 19 poorly differentiated endometrioid endometrial carcinomas (G3-EECs) compared with normal endometrium biopsies.
