Assay Background

Mast / stem cell growth factor receptor (SCFR), also known as the proto-oncogene c-Kit, tyrosine protein kinase Kit or CD117, is a protein encoded in humans by the KIT1 gene located on human chromosome 4 . Multiple transcription variants encoding different isoforms have been found for this gene2. KIT was first described by the German biochemist Axel Ullrich in 1987 as the cell homologue of the feline sarcoma viral oncogene v-kit3.

CD117 is a cytokine receptor (en) expressed on the surfaces of hematopoietic stem cells as other cell types. Altered forms of this receptor may be associated with certain types of cancer5. CD117 is a receptor with tyrosine kinase type III activity that binds Stem Cell Factor (a stem cell growth factor, belonging to the cytokine family), also known in English as “steel factor” or “c -Ligand kit “. When this receptor binds the stem cell factor (SCF), it then forms a protein dimer (en) that activates its intrinsic protein kinase activity, which in turn phosphorylates and activates signal transduction molecules that propagate this signal into the cell. . CD117 signal transduction plays a role in cell survival, proliferation and differentiation.

Assay Principle

The CD117 Kit is a solid phase sandwich Enzyme Linked-Immuno- Sorbent Assay (ELISA). A monoclonal antibody specific for CD117 has been coated onto the wells of the microtiter strips provided. Samples, including standards of known CD117 concentrations and unknowns are pipetted into these wells. During the first incubation, the CD117 antigen and a biotinylated monoclonal antibody specific for CD117 are simultaneously incubated.  After washing, the enzyme (streptavidin-peroxydase) is added. After incubation and washing to remove all the unbound enzyme, a substrate solution is added to induce a coloured reaction product. The intensity of this coloured product is directly proportional to the concentration of CD117 present in the samples.

  1. Add 100μl of each standard, sample, control and zero (Standard Diluent) in duplicate to appropriate number of wells
  2. Add 50μl of diluted biotinylated anti-CD117 to all wells
  3. Cover with a plastic plate cover and incubate at room temperature (18 to 25°C) for 1 hour
  4. Remove the cover and wash the plate
  5. Add 100μl of Streptavidin-HRP solution into all wells
  6. Cover with a plastic plate cover and incubate at room temperature (18 to 25°C) for 30 min
  7. Repeat wash step 4.
  8. Add 100μl of ready-to-use TMB Substrate Solution into all wells
  9. Incubate in the dark for 10-20 minutes* at room temperature. Avoid direct exposure to light by wrapping the plate in aluminium foil
  10. Add 100μl of H2SO4:Stop Reagent into all wells
