Human eye epithelium cells were stably transfected with KillerRed-Membrane to establish a cell line with bright red fluorescent cellular membranes, while no fluorescence is found in the cytoplasm. As a result, the plasma membrane with all its protrusions becomes clearly visible.
KillerRed is the first genetically encoded photosensitizer. It has been developed on the basis of the Anthomedusae chromoprotein anm2CP. Unlike chemical photosensitizers, KillerRed can be directly expressed by target cells, both individually and infusion with a target protein. It shows no cell toxicity before light activation. Upon green light irradiation, KillerRed generates reactive oxygen species (ROS) that damage the neighboring molecules. KillerRed phototoxicity is induced by green light irradiation at 540-580 nm and depends on light intensity, irradiation time and KillerRed concentration.
