The Rat Classical Complement Pathway ELISA Kit is for the qualitative/semi-quantitative determination of activation of the classical pathway of the complement system in rat serum and plasma samples. The Rat Classical Complement Pathway ELISA Kit is for research use only and not to be used for diagnostic procedures.

Rat Classical Complement Pathway Assay Kit

The complement system is an important component of the innate immune response and contributes to the first line of defense against foreign pathogens. It consists of a complex network of proteins and receptors found in the circulation, tissues, and other body fluids and also interacts with the adaptive immune system. Complement activation occurs through three pathways: the classical, lectin, and alternative pathways, which converge on activation of the central complement component C3 and ultimately lead to formation of terminal complement components, including the C5b-9 terminal complement complex (TCC). The classical pathway is primarily initiated when C1q binds to antibodies within immune complexes, triggering a cascade of proteolytic reactions that results in complement activation. The alternative pathway can also amplify complement activation initiated through the other pathways.

In research settings, assessment of classical complement pathway activity is used to investigate innate immune responses, immune complex-mediated processes, inflammation, and complement-related disease mechanisms. Researchers can evaluate pathway activity in experimental models of autoimmune and inflammatory conditions and investigate how alterations in complement components affect immune responses. Classical pathway activity can also be studied alongside the lectin and alternative pathways to characterize pathway-specific complement activation and identify potential defects or deficiencies in the complement system.

In clinical research, abnormalities in complement pathway activity have been investigated in a range of immune-mediated and infectious conditions. Deficiencies or functional defects affecting classical pathway components can impair immune complex clearance and increase susceptibility to certain infections or autoimmune manifestations. Measuring classical pathway activity, together with assessments of the lectin and alternative pathways and individual complement components, can help researchers characterize complement function and investigate pathway-specific abnormalities. ELISA-based pathway assays provide a convenient approach for evaluating classical complement activation in experimental samples.
