Luteinizing hormone (LH) is a glycoprotein hormone produced by the anterior pituitary gland and plays a critical role in the regulation of the reproductive system. Structurally, LH consists of two subunits—α and β. While the α subunit is common among other pituitary hormones such as follicle-stimulating hormone (FSH), thyroid-stimulating hormone (TSH), and human chorionic gonadotropin (hCG), the β subunit is unique and confers biological specificity to LH. The release of LH is stimulated by gonadotropin-releasing hormone (GnRH) from the hypothalamus, and it is essential for normal reproductive function in both males and females.
In men, LH acts directly on the Leydig cells of the testes to stimulate the production of androgens, primarily testosterone and dihydrotestosterone, which are crucial for the development of male secondary sexual characteristics and the maintenance of spermatogenesis. In clinical and research settings, LH is frequently measured to assess pituitary function, diagnose reproductive disorders, evaluate fertility status, and monitor gonadal function. Abnormal LH levels can indicate conditions such as hypogonadism, polycystic ovary syndrome (PCOS), pituitary tumors, or primary gonadal failure.
The hLH ELISA is a highly sensitive two-step sandwich immunoassay designed for the quantitative determination of LH in serum or plasma. The assay uses two specific monoclonal antibodies: one immobilized on the microplate to capture LH, and the other conjugated to horseradish peroxidase (HRP) to enable detection. Following the formation of a sandwich complex, a TMB substrate is added, producing a colorimetric signal directly proportional to the amount of LH in the sample. Absorbance is measured at 450 nm, and results are calculated using a standard calibrator curve, providing reliable and precise hormone level assessments for both diagnostic and research applications.
