NUCB2 encodes a multifunctional calcium/DNA-binding protein (396 amino acids in humans) that contains EF-hand motifs, a leucine-zipper region, and a signal peptide. The protein is processed by prohormone convertases into three peptide fragments: nesfatin-1, nesfatin-2 and nesfatin-3, of which nesfatin-1 is the most studied for its role in appetite regulation and energy homeostasis. NUCB2 (and the nesfatin-1 fragment) is expressed across many tissues (central nervous system, adipose tissue, pancreas, etc.). In non-disease settings, NUCB2/nesfatin-1 has been implicated in regulation of feeding behaviour, metabolic homeostasis, reproductive axis function and possibly stress responses. In the context of biomarker biology, NUCB2 has emerged in numerous studies of malignancy and metabolic disease. Elevated or dysregulated NUCB2 expression has been associated with worse prognosis in various cancers (breast, glioblastoma, renal cell carcinoma, hepatocellular carcinoma) and may correlate with proliferation/migration/invasion phenotypes in tumour cell lines.
