Anti–Endothelin Receptor A (ETAR) IgG antibodies are autoantibodies directed against the endothelin type A receptor, a G-protein–coupled receptor involved in vasoconstriction, inflammation, and fibrosis. Endothelin-1, the receptor’s primary ligand, plays a major role in vascular homeostasis and remodeling. When IgG antibodies target ETAR, they can act as functional agonists, promoting excessive receptor activation and contributing to pathological processes such as vascular injury, endothelial dysfunction, and profibrotic signaling. Because of these mechanistic links, anti-ETAR antibodies have drawn attention as potential biomarkers and mediators in autoimmune and vascular diseases.

In research settings, anti-ETAR IgG is used to study pathways of vascular inflammation, fibrosis, and autoimmunity. Levels of these antibodies are often measured in diseases such as systemic sclerosis, transplant-related complications (e.g., antibody-mediated rejection), and certain cardiovascular disorders, helping investigators understand disease progression and the interplay between immune responses and endothelial signaling. Functional assays also allow researchers to explore whether these antibodies have direct biological activity on endothelial cells, offering insights into disease mechanisms and potential therapeutic targets.

Clinically, anti-ETAR IgG antibodies are being investigated as diagnostic and prognostic biomarkers. In systemic sclerosis, for example, higher levels of these antibodies have been associated with digital ulcers, pulmonary hypertension, and more severe vascular complications. In transplantation, they may help predict graft dysfunction or rejection risk when combined with other biomarkers. Although not yet universally standardized for routine diagnostics, testing for anti-ETAR IgG is increasingly used in specialized centers to support risk stratification and guide patient monitoring in diseases characterized by vascular or autoimmune pathology.
