A recent publication in Clinical Cancer Research revealed some
ongoing pancreatic cancer research identifying key cancer biomarkers for developing laboratory tests

Binding of the sphingolipid S1P to hTERT stabilizes telomerase at the nuclear periphery by allosterically mimicking protein phosphorylation

Study Summary:

In normal adult cells, the structure at the ends of chromosomes, called the telomere, becomes progressively shorter with each replication cycle. When telomeres get too short or damaged, the cell stops dividing and becomes senescent. The enzyme telomerase maintains the integrity of telomeres, and phosphorylation stabilizes the catalytic subunit (called TERT) of this enzyme. In both normal fibroblasts and lung cancer cells, Panneer Selvam et al. found that binding of the phospholipid S1P to TERT mimicked protein phosphorylation and prevented TERT degration. Disrupting the interaction between S1P and TERT by either depleting S1P or mutating the interaction site in TERT impaired telomere maintenance and promoted senescence in cultured cells and decreased the growth of lung cancer cell xenografts in mice. Researchers utilized Eagle Biosciences’Biotin ELISA Assay Kit to measure levels of Biotin in Biotinylated S1P–bound hTERT proteins.

Why Measure NT-proBNP?

The N-terminal prohormone of brain natriuretic peptide (NT-proBNP) is
a 76 amino acid N-terminal protein that is cleaved to release brain natriuretic
peptide. Both NTproBNP and BNP are secreted mainly by the left cardiac
ventricle as a result of pressure overload and wall stretch. Elevated NT-proBNP
levels indicate the presence of ongoing myocardial stress and potentially an
underlying, possibly unsuspected, cardiac disorder.  

NT-proBNP is the N-terminal 76 amino-acid
cleavage product of proBNP that is released from cardiac ventricles in response
to increased wall stress and volume overload. NT-proBNP has a longer half-life
in the circulation (120 minutes) compared with the C-terminal BNP peptide (20
minutes). Therefore, the measurement of NT-proBNP offers excellent stability in
serum/plasma for reliable results.

A recent study at Vanderbilt University focused on investigating the various metabolites of olives and their anticancer properties.  This research was based on previous metabolomic studies identifying potential candidates for topoisomerase II poisons (agents for anticancer therapy). Researchers tested several olive metabolites and successfully demonstrated that three of them enhanced DNA cleavage and acted as covalent topoisomerase II poisons. Can l-Cysteine and Vitamin D Rescue Vitamin D and Vitamin D Binding Protein Levels in Blood Plasma of African American Type 2 Diabetic Patients?

 

Study Summary:

Vitamin D deficiency and an a rapidly increasing rate of diabetes have
become a worldwide epidemic, particularly affecting African Americans. This study demonstrated that there was a significant positive correlation was observed between lower Vitamin D Binding Protein (VDBP) and vitamin D and l-cysteine and glutathione blood levels in African American diabetic
patients; raising glutathione status via l-cysteine supplementation
upregulated both VDBP and 
VDBP/vitamin D receptor in monocytes. VDBP is a
key determinant of vitamin D blood levels. This novel finding will
impact the clinical practice of vitamin D supplementation and the
development of new therapies based on the potential of l-cysteine coupled with lower vitamin D doses to improve the efficacy and
levels of vitamin D and health outcomes in African Americans. Researchers utilized Eagle Biosciences’ 
25-OH Vitamin D ELISA to measure plasma levels of 25-OH Vitamin D.

 

Publication:

Subclinical Hypovitaminosis D and Osteoporosis in Breast Cancer Patients

 

Study Summary:

Vitamin D has also been reported to have anticancer activities against many cancer types, including breast cancer. The breakthrough that breast epithelial cells can locally manufacture active vitamin D from circulating precursors, makes the effect of vitamin D in breast cancer biologically conceivable. This study focuses on evaluating the circulating concentration of 25-hydroxy vitamin D and the bone mineral density status of breast cancer patients and to study their relation to the treatment received and the stage of breast cancer.  Researchers utilized Eagle Biosciences’ 25-OH Vitamin D ELISA to measure serum levels of 25-OH Vitamin D.

This study did in fact support the findings of increased incidence of hypovitaminosis D, osteoporosis and osteopenia in breast cancer patients. Therefore, it is concluded that there is value and importance of offering calcium and vitamin D supplements to breast cancer patients. It is recommended that breast cancer patients have a DXA scan on a yearly basis.

Abundant rodent furan-derived urinary metabolites are associated with tobacco smoke exposure in humans

Study Summary:

Furan, a possible human carcinogen, is found in heat treated foods and tobacco smoke. Past studies have shown that humans are capable of converting furan to its reactive metabolite, cis-2-butene-1,4-dial (BDA), and thus may be susceptible to furan toxicity. Human risk assessment of furan exposure has been hindered because of the lack of mechanism-based exposure biomarkers.

In order to investigate this further, a study was performed at the University of Minnesota for evaluate six furan metabolites by measuring their levels in urine from furan-exposed rodents as well as in human urine from smokers and nonsmokers. The targets of this study were the metabolites that result from direct reaction of BDA with lysine (BDA-N(α)-acetyllysine) and from cysteine-BDA-lysine cross-links (N-acetylcyteine-BDA-lysine, N-acetylcysteine-BDA-Nα-acetyllysine and their sulfoxides. Researchers utilize Eagle Bioscience’s Creatinine Microplate Assay to measure creatinine in urine samples.

Five of the six metabolites were identified in urine from rodents treated with furan by gavage. BDA-N(α)-acetyllysine, N-acetylcysteine-BDA-lysine and its sulfoxide were detected in most human urine samples from three different groups. The levels of N-acetylcysteine-BDA-lysine sulfoxide were more than 10 times higher than the corresponding sulfide in many samples. The amount of this metabolite was higher in smokers relative to non-smokers and was significantly reduced following smoking cessation. Our results indicate a strong relationship between BDA-derived metabolites and smoking. Future studies will determine if levels of these biomarkers are associated with adverse health effects in humans.

Mercapturic Acids Derived from the Toxicants Acrolein and Crotonaldehyde in the Urine of Cigarette Smokers from Five Ethnic Groups with Differing Risks for Lung Cancer

Study Summary:

Lung cancer is the leading cause of cancer death in the world and in order to have a better understanding of lung cancer susceptibility the different risks among smokers in varied ethnic groups needed to be evaluated. 

The results of the Multiethnic Cohort epidemiology study has clearly demonstrated that, compared to Whites and for the same number of cigarettes smoked, African Americans and Native Hawaiians have a higher risk for lung cancer whereas Latinos and Japanese Americans have a lower risk.  In addition, it was found that Acrolein and crotonaldehyde are two important constituents of cigarette smoke which have well documented toxic effects and could play a role in lung cancer etiology. 

Their urinary metabolites 3-hydroxypropylmercapturic acid (3-HPMA) and 3-hydroxy-1-methylpropylmercapturic acid (HMPMA), respectively, are validated biomarkers of acrolein and crotonaldehyde exposure.The levels of 3-HPMA and HMPMA were quanified in the urine of more than 2200 smokers from these five ethnic groups, and also carried out a genome wide association study using blood samples from these subjects. Creatinine levels were analyzed using Eagle Bioscience’s Creatinine Microplate Assay to measure creatinine in urine samples.

In conclusion, the overall results of this study suggest that acrolein and crotonaldehyde may be involved in lung cancer etiology, and that their divergent levels may partially explain the differing risks of Native Hawaiian and Latino smokers. No strong signals were associated with 3-HPMA in the genome wide association study, suggesting that formation of the glutathione conjugate of acrolein is mainly non-enzymatic, while the top significant association with HMPMA was located on chromosome 12 near the TBX3 gene, but its relationship to HMPMA excretion is not clear.
