Mouse Lectin Complement Pathway ELISA Kit

The complement system is an important component of the innate immune response and contributes to the first line of defense against foreign pathogens. It consists of a complex network of proteins and receptors found in the circulation, tissues, and other body fluids and also interacts with the adaptive immune system. Complement activation occurs through three pathways: the classical, lectin, and alternative pathways, which converge on activation of the central complement component C3 and ultimately lead to formation of terminal complement components, including the C5b-9 terminal complement complex (TCC). The lectin pathway is activated when pattern-recognition molecules, including mannose-binding lectin (MBL) and ficolins, recognize specific carbohydrate structures on the surface of pathogens or damaged cells. This initiates a proteolytic cascade involving complement components C4 and C2, leading to formation of the lectin pathway C3 convertase and subsequent complement activation.

In research settings, assessment of lectin complement pathway activity is used to investigate innate immune responses, pathogen recognition, inflammation, and complement-related disease mechanisms. Researchers can evaluate lectin pathway activity in experimental models of infectious, autoimmune, and inflammatory conditions and investigate how alterations in pathway components affect complement activation. Lectin pathway activity can also be assessed alongside the classical and alternative pathways to characterize pathway-specific complement responses and investigate complement deficiencies or functional abnormalities.

In clinical research, alterations in lectin pathway components and activity have been investigated in infectious, inflammatory, and immune-mediated conditions. Deficiencies or functional abnormalities involving lectin pathway components, such as MBL or ficolins, may influence susceptibility to infection and other complement-related processes. Measuring lectin pathway activity, together with assessments of the classical and alternative pathways and individual complement components, can help researchers characterize complement function and investigate pathway-specific abnormalities. ELISA-based pathway assays provide a convenient approach for evaluating lectin complement pathway activation in experimental mouse serum samples.
