Progesterone is a C-21 steroid hormone that plays a critical role in female reproductive physiology. Secreted in low amounts during the follicular phase of the menstrual cycle, progesterone levels begin to rise at the time of the luteinizing hormone (LH) surge and peak approximately 3 to 4 days afterward. These elevated levels persist for about 10 to 12 days following the LH peak before returning to baseline. Progesterone is essential for preparing the endometrium for embryo implantation and maintaining early pregnancy, making it a key biomarker in reproductive health.

In clinical and research settings, progesterone is widely used to assess ovulatory function and monitor early pregnancy. Measuring serum progesterone can confirm whether ovulation has occurred and is also valuable in evaluating luteal phase defects, predicting miscarriage risk, and diagnosing ectopic pregnancy. Progesterone levels are also important in fertility evaluations and are monitored during assisted reproductive treatments. However, values can be influenced by medications such as oral contraceptives, estrogen replacement therapies, ovulation-inducing drugs, and GnRH analogues. Surgical removal of the ovaries or chemotherapy can also significantly impact circulating progesterone concentrations.

The Progesterone ELISA Assay Kit provides a sensitive and specific method for quantifying progesterone levels in serum. This competitive immunoassay involves competition between endogenous progesterone and an HRP-labeled progesterone conjugate for limited antibody binding sites on a microplate. After washing to remove unbound materials, a TMB substrate is added, producing a color change that is inversely proportional to the progesterone concentration. The reaction is halted with a stopping solution, and absorbance is measured at 450 nm. A standard curve is then used to determine progesterone levels in test samples, offering valuable data for both diagnostic and investigational reproductive applications.
