Monomethyl auristatin E (MMAE) is a potent microtubule-disrupting agent used as the cytotoxic payload in many antibody–drug conjugates (ADCs), such as brentuximab vedotin. In these ADCs, MMAE is linked to a monoclonal antibody via a cleavable linker that releases the active drug inside target cells upon internalization and lysosomal degradation. The term “intact MMAE ADC” refers to the full, conjugated form of the ADC prior to linker cleavage—comprising both the antibody and the attached MMAE molecules.
