Anti-deamidated gliadin peptides (anti-DGP) IgA are autoantibodies that target gliadin peptides modified by tissue transglutaminase through deamidation, a process that increases their immunogenicity. These antibodies are more specific and sensitive than traditional anti-gliadin antibodies (AGA) and are primarily associated with celiac disease, an autoimmune condition triggered by gluten ingestion in genetically predisposed individuals. The deamidation of gliadin enhances its binding to HLA-DQ2 or DQ8 molecules, which in turn stimulates an abnormal immune response, including the production of anti-DGP antibodies.
In clinical settings, anti-DGP IgA is used as part of the serologic workup for celiac disease, especially in cases where anti-tTG IgA results are inconclusive or when additional confirmation is needed. It is particularly useful in younger children under the age of two, who may not yet produce high levels of anti-tTG IgA. While less commonly used as a standalone test, anti-DGP IgA can help support diagnosis, assess dietary compliance, and monitor immune response to gluten. In research, anti-DGP IgA serves as a tool for exploring the early stages of celiac disease pathogenesis, understanding mucosal immune activation, and evaluating the effectiveness of new diagnostic markers or gluten-free treatment approaches in various populations.
