PMN-Elastase is a 30 kDa glycoprotein and a member of the serine protease family, released from the azurophil granules of neutrophil granulocytes in response to immune stimulation, cellular irritation, or disintegration. The PMN-Elastase Serum/Plasma ELISA uses a highly specific sandwich immunoassay format involving both polyclonal and monoclonal antibodies. The assay captures PMN-Elastase—including both its free and complexed forms—onto antibody-coated microplate wells. After sequential incubation with a monoclonal antibody and a peroxidase-labeled secondary conjugate, the resulting colorimetric reaction is measured at 450 nm. The color intensity is directly proportional to the concentration of PMN-Elastase, and quantification is achieved using a standard calibration curve. This dual-antibody system ensures high specificity and minimizes false results, providing a reliable tool for diagnosing and monitoring neutrophil-driven inflammation.
