The OxyStat Assay is a colorimetric test system designed to measure the total oxidative status or capacity (TOS/TOC) in EDTA plasma, serum, and other biological samples. This quick and straightforward assay quantifies total peroxide concentration, providing a reliable indicator of oxidative stress levels. The method is based on the reaction of biological peroxides with peroxidase, followed by a colorimetric reaction using tetramethylbenzidine (TMB) as a substrate. The resulting color intensity, measured photometrically at 450 nm, correlates directly with the concentration of circulating biological peroxides in the sample.
By assessing the relationship between free radicals and biological peroxides, the OxyStat assay enables the characterization of oxidative status in cells and tissues, which are highly sensitive to oxidative stress. If not neutralized by antioxidants, free radicals and reactive oxygen species produce organic peroxides and hydroperoxides—the earliest markers of oxidative damage. The OxyStat assay thus offers a valuable tool for studying these processes efficiently.
Unlike traditional methods such as HPLC or TBARS assays, which are either costly or detect only specific degradation products, the OxyStat assay provides a rapid, cost-effective, and comprehensive approach to evaluating oxidative stress.
