Osteonectin, also known as SPARC (Secreted Protein Acidic and Rich in Cysteine) or BM-40, is a 32.7 kDa calcium-binding glycoprotein primarily synthesized by osteoblasts, endothelial cells, and megakaryocytes. It plays a vital role in tissue remodeling, especially in the context of bone metabolism and endothelial cell proliferation and repair. Due to its involvement in both skeletal and vascular systems, osteonectin is considered a key biomarker in research focusing on bone formation, angiogenesis, and tissue regeneration.
In research and clinical contexts, osteonectin is studied as a marker of active bone remodeling and vascular repair processes. It is particularly valuable in investigating conditions such as osteoporosis, bone metastases, fracture healing, and cardiovascular disease. Its expression levels can provide insights into the dynamics of extracellular matrix remodeling, making it useful in both orthopedic and oncology research. Osteonectin’s role in tissue integrity and repair also makes it a candidate biomarker for studies involving wound healing and fibrotic diseases.
The Osteonectin ELISA is a competitive immunoassay designed for the quantitative determination of osteonectin levels in biological samples. In this assay, osteonectin in the sample competes with immobilized osteonectin on the microplate for binding to a specific antibody. Detection is carried out using a peroxidase-conjugated secondary antibody and tetramethylbenzidine (TMB) substrate. The enzymatic reaction is stopped with an acidic solution, and the color intensity—measured at 450 nm—is inversely proportional to the concentration of osteonectin in the sample. This assay enables sensitive and specific quantification for applications in bone, vascular, and regenerative biology research.
