Fetuin-A (alpha-2-Heremans-Schmid glycoprotein) is a liver-derived plasma glycoprotein that plays a multifaceted role in systemic physiology. It is known for its function as a negative acute-phase reactant and is involved in regulating inflammation, insulin signaling, and vascular calcification. Biochemically, Fetuin-A inhibits ectopic calcification by forming soluble complexes with calcium and phosphate, thus preventing mineral deposition in soft tissues. It also modulates insulin receptor activity and has been implicated in metabolic disorders such as type 2 diabetes and metabolic syndrome, where elevated levels often correlate with insulin resistance.
In both research and clinical settings, Fetuin-A is increasingly recognized as a biomarker for a range of pathological conditions. In cardiovascular disease, altered Fetuin-A levels have been associated with increased risk of atherosclerosis and arterial stiffness. In chronic kidney disease (CKD), low Fetuin-A levels are predictive of vascular calcification and poor outcomes. Additionally, in neurodegenerative and inflammatory diseases, Fetuin-A has been studied as a marker of systemic inflammation and blood-brain barrier dysfunction. Its broad involvement in metabolic, vascular, and inflammatory pathways makes it a promising candidate for biomarker discovery, therapeutic targeting, and risk stratification in both translational research and clinical diagnostics.
