Diamine oxidase (DAO) is an essential enzyme responsible for breaking down histamine, with its primary activity occurring in the intestine. When DAO levels are low or its activity is inhibited, histamine cannot be degraded efficiently, leading to symptoms of histamine intolerance. These symptoms may include headaches, gastrointestinal discomfort, nasal irritation, skin flushing, swelling, and allergy-like reactions after consuming histamine-rich foods.

In research and clinical settings, DAO serves as a useful biomarker for evaluating histamine metabolism and identifying potential histamine intolerance. Measuring DAO concentration—often alongside DAO enzymatic activity—can help differentiate histamine intolerance from other causes of similar symptoms and guide dietary or therapeutic interventions. It is particularly valuable for assessing patients with recurrent migraines, digestive complaints, food sensitivities, or chronic inflammatory conditions.

The DAO ELISA allows for quantitative determination of serum DAO levels and supports monitoring of histamine-related disorders, including tracking symptom improvement during a histamine-restricted diet.
