Ceruloplasmin is a copper-binding glycoprotein primarily synthesized in the liver, functioning as a ferroxidase that catalyzes the oxidation of ferrous iron (Fe²⁺) to ferric iron (Fe³⁺), which is essential for iron transport and homeostasis. Beyond its role in iron metabolism, ceruloplasmin acts as an acute-phase protein, increasing in response to inflammation, infection, or tissue injury, and exhibits antioxidant properties by scavenging free radicals. Abnormal ceruloplasmin levels are associated with disorders of copper metabolism, such as Wilson’s disease, and can reflect oxidative stress, liver dysfunction, and certain neurodegenerative diseases.
Ceruloplasmin ELISA (enzyme-linked immunosorbent assay) enables the quantitative measurement of ceruloplasmin in serum, plasma, or other biological fluids with high sensitivity and specificity. In research settings, ceruloplasmin ELISAs are used to study iron and copper metabolism, oxidative stress mechanisms, and the protein’s involvement in inflammatory processes, neurodegeneration, and cardiovascular disease. Researchers also use it to evaluate the effects of experimental drugs or interventions on oxidative stress and metal homeostasis.
In clinical settings, ceruloplasmin ELISA is valuable for diagnosing and monitoring disorders related to copper metabolism, including Wilson’s disease and aceruloplasminemia. It can also serve as a marker of systemic inflammation or liver function, helping clinicians assess disease progression or response to therapy. Additionally, ceruloplasmin levels can provide insight into oxidative stress status in patients with chronic diseases, guiding both diagnostic and therapeutic decisions.
