Vitamin B2 (riboflavin) is an essential nutrient involved in numerous metabolic processes, primarily through its biologically active derivatives—flavin mononucleotide (FMN) and flavin adenine dinucleotide (FAD). These flavin-nucleotides act as crucial coenzymes in the transport of hydrogen ions and electrons, supporting carbohydrate, fat, and protein metabolism. In circulation, most FMN and FAD are protein-bound, with only a small fraction found in free form. Because urinary riboflavin levels fluctuate significantly, serum measurement is preferred for accurate assessment of vitamin B2 status.

Clinically, vitamin B2 testing is indicated in conditions associated with malabsorption, increased metabolic demand, or poor nutritional intake, such as chronic diarrhea, preeclampsia, hypothyroidism, diabetes mellitus, alcoholism, anorexia, and lactose intolerance. Deficiency can impair energy production, antioxidant function, and tissue health, making riboflavin assessment valuable for diagnosing nutritional deficiencies and guiding supplementation strategies.

The Vitamin B2 test kit utilizes a microbiological assay in which vitamin B2–dependent growth of Lactobacillus rhamnosus is measured turbidimetrically after incubation. The degree of bacterial growth is directly proportional to the concentration of vitamin B2 in the sample, allowing accurate quantification via ELISA reader. This assay is suitable for human and veterinary diagnostics as well as research applications, supporting studies on nutrition, metabolic disorders, and clinical conditions affecting vitamin B2 status.
