Kappa light chains are one of the two types of immunoglobulin light chains produced by B cells, the other being lambda. These light chains pair with heavy chains to form complete antibodies, playing a central role in the adaptive immune response. Free kappa light chains, which are not bound to heavy chains, are normally present at low levels in serum and urine. Elevated levels can indicate abnormal B cell activity, including plasma cell disorders, autoimmune diseases, and chronic infections. Because of their small size and rapid clearance from circulation, kappa light chains can serve as sensitive biomarkers for monitoring immune dysregulation and pathological protein production.

The Kappa ELISA (enzyme-linked immunosorbent assay) enables precise quantification of free kappa light chains in serum, plasma, or urine. In research settings, it is widely used to study B cell function, immunoglobulin synthesis, and the dynamics of humoral immune responses. Researchers also use kappa ELISAs to investigate mechanisms of autoimmune disease, lymphoproliferative disorders, and multiple myeloma, as well as to evaluate the impact of therapeutic interventions on antibody production.

In clinical applications, measuring free kappa light chains provides important diagnostic and prognostic information. It is commonly used in conjunction with lambda light chain measurements to calculate the kappa/lambda ratio, which helps detect and monitor plasma cell disorders such as multiple myeloma and light chain amyloidosis. Additionally, elevated kappa levels can indicate abnormal B cell activity in autoimmune conditions or chronic inflammation. Serial measurements of kappa light chains can guide treatment decisions, monitor disease progression, and assess response to therapy, making it a valuable tool in both hematology and immunology practice.
